Accipitridae Sex Determination | Real-Time PCR Fluorescence | Lyophilized
The Eagle DNA Sex Identification Kit is a lyophilized real-time PCR fluorescence assay designed for molecular sex determination in eagles and relevant Accipitridae samples. The assay uses two sex-related detection targets monitored through the FAM and VIC fluorescence channels.
Supplied with One-Step SSB extraction reagent, the kit supports rapid crude DNA preparation from suitable feather, feather follicle, swab and tissue samples. The clarified extraction supernatant can be used directly as a PCR template, without an additional nucleic acid purification step. Both 48-test and 96-test formats are available.
Real-Time PCR Fluorescence Detection:Two sex-related detection targets monitored through FAM and VIC channels.
Lyophilized PCR Reagents:Convenient reconstitution format for routine laboratory testing.
One-Step SSB Sample Preparation:Rapid crude DNA extraction from suitable feathers, feather follicles, swabs and tissue samples.
Direct Use of Crude Extract:Clarified One-Step SSB supernatant can be added directly to the PCR reaction without an additional purification step.
Multiple Reaction Formats:Available in individual reaction tubes, 8-strip tubes and multi-reaction tube formats.
Flexible Testing Capacity:48-test and 96-test packaging options for laboratory testing and research applications.
Molecular sex identification is an important laboratory tool in avian research, wildlife management, conservation breeding and captive bird management. For eagles and relevant Accipitridae samples, DNA-based sex determination provides a molecular approach when external morphological characteristics are insufficient for reliable sex identification.
The Eagle DNA Sex Identification Kit uses specific primers and fluorescent probes for two sex-related nucleic acid targets. During real-time PCR amplification, the two targets generate fluorescence signals detected in the FAM and VIC channels. The amplification curves and Ct values are analyzed using a compatible real-time PCR instrument.
The kit combines lyophilized PCR reagents with a supplied One-Step SSB extraction reagent. For suitable feather, feather follicle, swab and tissue samples, the crude extraction supernatant can be used directly in the amplification reaction. This helps simplify sample preparation and reduces the need for a separate nucleic acid purification step in the specified workflow.
The assay contains specific primers, fluorescent probes, enzymes and reaction buffer components designed for the detection of two sex-related nucleic acid targets. During amplification, the probes generate fluorescence signals in the FAM and VIC channels, which are monitored in real time by the PCR instrument.
After amplification, the fluorescence curves are evaluated together with the positive and negative controls. The combination of FAM and VIC signals is used to interpret the sample result according to the kit's result interpretation rules.
FAM: Sex-related detection target 1
VIC: Sex-related detection target 2
The target identities are described by their fluorescence channels in this product overview. Please refer to the kit manual for detailed assay information and result interpretation.
The kit is designed for laboratory testing of suitable avian samples. The supplied One-Step SSB reagent can be used for rapid crude nucleic acid extraction from the following sample types:
Feathers with follicles: Approximately 1–2 feathers with intact follicles.
Swab samples: The swab tip containing collected cells.
Tissue samples: Approximately 40 mg of tissue, according to the manual.
The One-Step SSB procedure takes approximately 10 minutes. After sample mixing, room-temperature incubation and centrifugation, the clarified supernatant can be used directly as the PCR template. The recommended crude extract input is 5 μL per reaction.
EDTA-anticoagulated whole blood: Whole blood is listed as a sample type, but it should be processed according to the requirements of a genomic DNA extraction kit. The One-Step SSB crude extraction workflow described above should not be assumed to apply to whole blood.
Sample Collection:Collect a suitable feather with follicle, swab or tissue sample.
One-Step SSB Extraction:Add the sample to One-Step SSB reagent, mix, incubate and centrifuge according to the kit manual.
Template Preparation:Transfer 5 μL of clarified supernatant into the PCR reaction.
Lyophilized Reagent Reconstitution:Reconstitute the lyophilized PCR reagent according to the selected packaging format.
Real-Time PCR Amplification:Perform amplification using a compatible instrument with FAM and VIC detection channels.
Fluorescence Analysis:Review the amplification curves and controls, then interpret the result according to the FAM/VIC signal combination.
