Rapid Real-Time Fluorescence PCR

The kit uses a rapid PCR workflow with fluorescence acquisition during amplification.

The complete PCR program includes:

  • 55°C for 5      minutes

  • 95°C for 2      minutes

  • 40 cycles of:      

    • 95°C for 10       seconds

    • 58°C for 15       seconds with fluorescence acquisition

The recommended sample is a rectal swab sample.

Sample preparation is designed as a rapid processing workflow:

  1. Collect a      feline rectal swab using a flocked swab.

  2. Place the      swab tip into the lysis buffer.

  3. Vortex for 30      seconds.

  4. Transfer 20      μL of processed sample into the reconstitution solution.

  5. Vortex for 10      seconds.

  6. Use the      processed sample as the PCR template.

This procedure is based directly on the PC503 product instructions.

Step 1 – Sample Collection

Collect a feline rectal swab sample.

Step 2 – Rapid Sample Processing

Place the swab into lysis buffer and vortex for 30 seconds.

Step 3 – Reconstitution

Transfer 20 μL processed sample into the reconstitution solution.

Step 4 – PCR Setup

Transfer 20 μL processed sample into the lyophilized PCR reagent.

Step 5 – Real-Time Fluorescence PCR

Run the PC503 PCR program and collect fluorescence signals.

Step 6 – Result Analysis

Analyze amplification curves and Ct values.

Result Interpretation

Ct Value Analysis

Results are interpreted according to the real-time PCR amplification curves and Ct values.

  • Ct ≤ 36.0:      Positive

  • Ct > 36.0:      Negative

The threshold should be adjusted according to the amplification curve, negative control performance, and optimal linearity.

A clear S-shaped amplification curve above the threshold is expected for a positive reaction.
